Abstract
Common bean (Phaseolus vulgaris L.) is the most important legume crop for small farmers worldwide. Weeds, through competition, cause yield reductions in common bean. Through genetic engineering, herbicide resistant lines have been developed in a number of species, e.g. glufosinate (Liberty®) resistance. Common bean transformation has been pursued using biolistic and Agrobacterium methods with little success. The objectives of this study were to develop a more efficient transformation method for common bean. The study utilized two varieties, Tio Canela 75 and Morales. Four pollen germination media were evaluated and both sucrose and calcium concentrations were varied. To determine the optimum voltage for the introduction of the transgene, three dyes (trypan blue, cotton blue, and aniline blue) and the gfp gene were evaluated. Two methods of electroporation, direct electroporation with an electrode and using cuvettes, and three methods of pollination (‘paper,’ ‘agarose,’ and ‘droplet’) were evaluated. The putative transformants were evaluated genotypically, with PCR and Southern Blot, and phenotypically, with glufosinate at 100, 500, and 800 g GA/ha. The results indicate that one of the liquid pollen germination media was superior and that the 10% sucrose concentration gave the best results. Variation in calcium concentration did not have a significant effect on germination. The trypan blue dye, used to determine the best electroporation conditions, was optimized at 0.04% concentration. The selected transformation conditions were 4.0 kV/cm, 50 μF and 350 Ω. In total, 1,108 pollinations were completed, 667 with Morales, and 441 with Tio Canela 75, resulting in 80 and 14 pods, respectively. ‘Droplet’ was the superior pollination method and the ‘cuvette’ method was the best electroporation method. After evaluation with PCR, Southern Blot, and glufosinate, it was determined that transformation was not achieved. El frijol común (Phaseolus vulgaris) es la leguminosa más importante para pequeños agricultores en países en vías de desarrollo y desarrollados. Las malezas provocan competencia y disminución en rendimiento del frijol. Mediante ingeniería genética se han obtenido plantas resistentes a herbicidas en otros cultivos, con el transgen que confiere resistencia a glufosinato (Liberty®). La trasformación de frijol se ha experimentado con biobalística y Agrobacteria con poco éxito. Los objetivos de este estudio fue desarrollar un método más eficiente de transformación. El estudio utilizó las variedades de frijol ‘Tío Canela-75’ y ‘Morales’. Se evaluaron cuatro medios de germinación en los que se variaron la sucrosa y el calcio. Para determinar el voltaje adecuado para introducir el ADN, se evaluaron tres tintes (trypan blue, cotton blue y aniline blue) y el gen gfp. Se evaluaron dos métodos de electroporación cubeta y electrodo; y tres métodos de polinización ‘papel’, ‘agarosa’ y ‘gota’. Los transformantes putativos se evaluaron genotípicamente (PCR y Southern Blot) y fenotípicamente, con aplicación de glufosinato a 100, 500 y 800 g GA/ha. Se encontró que un medio líquido es superior para germinación de polen, con una concentración de sucrosa de 10%. La variación en concentración de calcio no tuvo efecto significativo en la germinación de polen. El tinte trypan blue funcionó adecuadamente a una concentración de 0.04%. Para determinar el voltaje adecuado para electroporación, las condiciones de electroporación seleccionado después de evaluación fueron de 4.0 kV/cm, 50 μF y 350 Ω. Se realizaron 1,108 polinizaciones, 667 con Morales y 441 con Tío Canela 75, resultando fecundadas 80 de Morales y 14 de Tío Canela 75. El método de polinización mejor fue de ‘gota’ y de electroporación fue de ‘cubeta’. Después de evaluación con PCR, Southern Blot, y glufosinato, se determinó que no hubo transformación genética.
